چکیده

Sialylated Asn-linked oligosaccharide units that are present in glycoproteins are often extensively heterogeneous in number, in the linkage type and location of the sialic acid (NeuAc) residues, and in the underlying asialo core oligosaccharides. Their structural characterization requires the complex mixture of oligosaccharides to be separated and purified to homogeneity. Baenziger and coworkers1~4) have introduced some rapid and sensitive methods which involve reducing the oligosaccharides with NaB3H4 and subsequently fractionating the mixture by HPLC. Recently, Yamamoto et a/.5 6) have developed a fluorescent HPLCprocedure for a sensitive detection and fractionation of anionic oligosaccharides. As an alternative to radioactive and fluorescent HPLC,procedures for labeling oligosaccharides with the ultra-violet absorbing reagent, /?-aminobenzoic acid ethyl ester (ABEE), have

Determination of monosaccharides in cider by reversed-phase liquid chromatography

A reversed-phase high-performance liquid chromatographic (HPLC) method is described for the simultaneous determination of aldoses and uronic acids in cider, previously derivatized with p-aminobenzoic ethyl ester (ABEE). Narrow-bore C8 columns are recommended as this alternative provides good separation efficiency, along with greater economy and sensitivity. Detection limits for aldoses (glucose...

SEPARATION OF NONHISTONE HIGH MOBILITY GROUP (HMG) FROM HUMAN LYMPHOCYTES BY HIGH PERFORMANCE LIQUID CHROMATOGRAPHY

The high mobility group (HMG) of nonhistone proteins have been investigated using two high performance liquid chromatographic techniques (HPLC). Reversed-phase HPLC under conditions of 50 mM triethylamine adjusted to pH 2.2 with phosphoric acid (solvent A) and 95% acetonitrile in water (solvent B) was used to separate proteins primarily on the basis of differences in the overall hydrophobi...

Separation of Sedative – Hypnotic Drugs with Mixed Micellar Liquid Chromatography

Separation of ten sedative- hypnotic drugs was performed by RP-HPLC using mixed micellar mobile phase. Effect of temperature, type and amount of organic modifier in mobile phase on efficiency (N) and asymmetry factor (B/A) showed that, the appropriate conditions for a good separation were 35°C and 7% (V/V) butanol in mobile phase.  Variations of selectivity factor versus butanol concent...

Determination of some B Vitamins in Sour Cherry Juice Using Dispersive Liquid-liquid Microextraction Followed by High-performance Liquid Chromatography

Dispersive liquid-liquid microextraction method (DLLME) combined with high-performance liquid chromatography-ultraviolet detection (HPLC-UV) was used to determine thiamine (B1), nicotinamide (B3) and pyridoxine (B6) in sour cherry juice. This method was rapid, simple and sensitive. Separation was accomplished using a C18 column. The optimum chromatographic conditions were found to be: mobile ph...

Simplified oral pancreatic function test.

The standard Bentiromide test and a new modified test using p-aminosalicylic acid (PAS) as a pharmacokinetic marker for p-aminobenzoic acid (PABA) have been evaluated in the detection of pancreatic exocrine insufficiency in children. The conventional two day test using a colorimetric assay for urinary PABA discriminated poorly between five children with pancreatic insufficiency and 13 others wi...
